#25 Caloric Restriction Deep-Dive & Longevity Secrets of Long-Lived Animals | Dr. Steven Austad

All platforms / Episode Transcript: https://livelongerworld.substack.com/p/saustad

Dr. Steven Austad is a professor in the Healthy Aging department at the University of Alabama and also scientific director of the American Federation for Aging research. He has a fascinating entry into the world of aging research - it all began with training lions!

There are many open questions on the caloric restriction research for enhancing longevity and we dive into that. Further, a lot of Dr. Austad’s work focuses on what long-lived animal species can teach us about living longer and healthier lives. His lab works on exotic species, like clams that live more than 500 years, and hydra that don’t age at all, in order to discover aging treatments. He also tries to uncover the mysteries around why men and women respond differently to longevity therapeutics.

#22 The Cancer Diet Connection & Otto Warburg - Nobel Prize Scientist under Hitler | Sam Apple, Author of Ravenous

All platforms / Episode Show Notes: https://livelongerworld.substack.com/p/sapple

Sam Apple is the author of Ravenous: Otto Warburg, The Nazis, and the Search for the Cancer-Diet Connection.

Otto Warburg was a Nobel-Prize winning German scientist during the era of Hitler and the Nazi’s and he was the first person to put the metabolic story of cancer on the map. The famous “Warburg Effect” that shows how cancer cells have an increase in glucose uptake even in the presence of oxygen, is named after him.

It was a fascinating conversation talking about how the story of cancer research has evolved - how cancer cells eat to the discovery of oncogenes to coming full circle and making the connection between the two, and what the debate still holds in the story. We also talk about how cancer cells take up a lot of glucose, feed on sugar, and the connection between obesity and cancer. I hope you enjoy the conversation!

#21 Selenium's Dual Role in Health & Aging in the Womb | Dr. Vadim Gladyshev

All platforms / Episode Show Notes: https://livelongerworld.substack.com/p/vgladyshev

Dr. Vadim Gladyshev is a professor at Harvard Medical school and a Director of redox medicine at Brigham and Women’s hospital, where he runs his own lab.  We had a brilliant discussion which started off by talking about his work on selenium. Selenium is found is Brazil nuts and we discussed its connection to aging and cancer prevention, and also some of the harmful effects it can have. Dr. Gladyshev calls it the good, the bad, the ugly nature of selenium.

Then, we discussed a fair bit on his theory of aging before and after development. Most people believe that the germline which is the reproductive cells do not age, but his lab’s work has shown how the germline does in fact age. On this point, his lab also has interesting results on how rapamycin during development extended lifespan in mice.

It’s important to understand the basic science and understand more on the aging process in order to crack it. Dr. Gladyshev shares his views on this too and his research is certainly a step towards cracking the aging code. I hope you enjoy the conversation!

Dr. Michael Levin is an American developmental and synthetic biologist at Tufts University where he heads his own lab. He also directs the Tufts Center for Regenerative and Developmental Biology.

His groundbreaking research focuses on bioelectrical signals that dictate the collective intelligence of cells and direct it to form tissues and organs. His work on bioelectric signals has important applications for how we understand cancer, regenerative medicine and even aging. In fact, he is collaborating with David Sinclair to see how his work can be applied to reverse aging.

Michael Levin’s work is fascinating and will surely leave you thinking. I hope you enjoy the conversation!

#18 Longevity Supplements - Interventions Testing Program Results | Dr. Richard Miller

#18 Longevity Supplements - Interventions Testing Program Results | Dr. Richard Miller

Dr. Richard Miller runs one of the labs for the Interventions Testing Program or the ITP, which is largely considered the gold standard for testing longevity drugs. This was one of the best conversations I’ve had on longevity supplements that affect lifespan and those that might me subject to mere hype. Dr. Miller is rigorous in his approach and states the data clearly. He also has opinions on lifespan vs. healthspan that are unique from those of most others and worth paying attention to. I genuinely walked away having learned so much from this conversation and feeling more awakened, and I hope it is useful to you too in parsing the data out there on longevity supplements.

In today’s episode, you will learn:

- The 4 winning drugs that increase lifespan in mice as tested by the ITP: Rapamycin, Acarbose, 17-alpha estradiol, Canagliflozin

- 3 other drugs that produce small lifespan benefits: NDGA, Glycine, Captopril

- Drug combinations: Rapamycin + Acarbose, Rapamycin + Metformin

- Drugs that show no lifespan increase in mice in the ITP: Resveratrol, Nicotinamide-Riboside (NR), Fisetin, Curcumin, MCT Oil, Green tea extract, Fish Oil + more

- Richard’s thoughts on lifespan vs. healthspan

#17 Epigenetic Reprogramming, Yamanaka Factors, Cryonics & COVID Lab Leak | Yuri Deigin

Yuri Deigin is the founder and CEO of YouthBio Therapeutics which is a company that is working on Epigenetic reprogramming or using the Yamanka factors to achieve partial reprogramming.

Epigenetic reprogramming has been in the news lately because of several companies being started in the space, with some notable ones being Altos Labs funded by Jeff Bezos and NewLimit founded by Brian Armstrong, CEO of Coinbase. If you’re not familiar with this trending, hyped (rightly so this time - it is pretty amazing!) research on Epigenetic reprogramming and Yamanaka factors to turn cells back to a younger state, we dive into it in today’s episode!

As a last point to Yuri’s introduction, because I’d be remiss if I didn’t mention it, you may have heard of his name recently because he was in fact one of the first people to talk about the COVID lab leak theory and wrote extensively on it. This speaks to his scientific mindset and desire to uncover the truth.

In today’s episode, you will learn:

- The Strong Epigenetic Theory of Aging, as coined by Yuri (He calls David Sinclair’s theory The Weak Epigenetic Theory of Aging)

- Epigenetic Reprogramming using Yamanaka factors to turn the cell into a younger state

- What is it and what are the open questions in the field

- What are the cancer risks associated with it

- Why Yuri believes in Cryonics

- Why Yuri got interested in uncovering the COVID-19 Lab Leak theory

- Pharma in Russia

#13 Dr. David Gems | Hyperfunction & Programmatic Theory of Aging and Critique of the Hallmarks of Aging

#13 Dr. David Gems | Hyperfunction & Programmatic Theory of Aging and Critique of the Hallmarks of Aging

My guest today is Dr. David Gems who runs the C. Elegans Aging lab at the University College London. We had a fascinating discussion talking about emerging paradigms in the theories of aging. We discussed the hyperfunction theory or the programmatic theory of aging which gives a better integrated understanding of the different theories of aging that have been presented such as damage accumulation, evolutionary, antagonistic pleiotropy. It has been assumed that molecular damage is always the cause of aging, but the programmatic theory of aging shows that that might not be the case. We also discussed his critique of the hallmarks of aging and how there should be a better paradigm for the aging field. Lastly, we also discussed how research from his lab on C. Elegans might explain some of the conflict around whether sirtuins affect lifespan and how C. Elegans also show versions of this programmatic theory of aging, which can give us a better understanding of how to study aging.

Note: There is a point in the conversation (40:41) where David pulls up some slides to better explain the Multifactorial Model of Aging. So if you are listening to the audio version, you might want to refer to YouTube or the show notes.

#12 Dr. Bruce Ames | Longevity Micronutrients, Triage Theory, and Vitamin D for Healthy Aging

#12 Dr. Bruce Ames | Longevity Micronutrients, Triage Theory, and Vitamin D for Healthy Aging

My guest today is Dr. Bruce Ames who has received several awards and accolades for developing the Ames test which is used to detect carcinogens in commercial products. He showed how there were carcinogens in products such as hair dyes and children’s pajamas!

Later, Dr. Bruce Ames turned to studying micronutrients and how deficiency of important vitamins and minerals leads to accelerated aging. He came up with the Micronutrient triage theory, which says that when nutrient intake is insufficient, the body has a rationing mechanism and favors proteins needed for immediate survival at the expense of protection against future damage.

Dr. Bruce Ames is 93 years old and says that he is still exercising and eating healthy greens cooked by his lovely Italian wife! However, he says his cognitive capacity has slowed down (this is why we need rejuvenation therapies, even for the brain: listen to my episode with Dr. Jean Hebert who talks about Brain Aging and his strategy to reverse aging through replacement of the body parts: https://youtu.be/oerGUepWsH8).

I am grateful that I had a chance to converse with him on Vitamin D, longevity micronutrients, how micronutrient deficiency can lead to accelerated aging, his take on the science behind reversing aging, and his advice to younger people.

This is a shorter episode and I hope you learn more about the importance of micronutrients!

#10 Dr. Collin Ewald | Collagen Protein & Extracellular Matrix During Aging

#10 Dr. Collin Ewald | Collagen Protein & Extracellular Matrix During Aging

My guest today is Dr. Collin Ewald who is a professor at the Health Sciences department at ETH Zurich. His lab focuses on understanding the extracellular matrix (ECM) and how it changes with age. Collin explains the role of the ECM in tissue repair, muscle strength, and overall longevity. In fact, Collin’s lab is also working on establishing the ECM as a biomarker for health and disease risk. So, studying the ECM can indeed tell us a lot about the aging process and ways to intervene.

Think of the ECM as the environment that surrounds the cell or the proteins secreted from the cell. It holds our organs and tissues in place. You’ve probably heard of collagen protein - it is largely found in the ECM.

Why is the ECM relevant for longevity research? Turns out, the ECM changes with age. In fact, each cell has a unique ECM and cancer cells can be detected by their ECM composition type! There is also a connection between the ECM and the muscle, and damage to collagen with age results in weakening of our muscles.

We discuss changes in collagen with age, how sugar impacts collagen, cancer cells & ECM, the overexpression of certain collagens with age, changes in collagen causing muscle impairment with age, and longevity interventions his lab is studying to repair the impairment damage.

(This podcast can be a little technical, especially in the beginning, so I’d recommend referring to the show notes for better understanding. The video version of the podcast also contains some explanations on the video slide itself.)

#09 Dr. Irina & Michael Conboy | Tissue Rejuvenation Via Plasma Dilution, Neutral Blood Exchange and Parabiosis

#09 Dr. Irina & Michael Conboy | Tissue Rejuvenation Via Plasma Dilution, Neutral Blood Exchange and Parabiosis

My guests in today's episode are Dr. Irina and Michael Conboy of the Department of Bioengineering at the University of California Berkeley.

Their groundbreaking research focuses on tissue rejuvenation via plasma dilution and heterochronic parabiosis, where the Conboys took a young mouse and an older mouse and sutured them together, such that the animals shared blood and environment. The old mice became rejuvenated & younger. They’ve also pioneered a process called the Neutral Blood Exchange or Therapeutic Blood Exchange, through which dilution of old blood plasma can lead to true rejuvenation in the brain & other tissues of mice to a younger state!!

Further, we touch upon Irina's work showing how decreased Oxytocin levels with age contributes to poor muscle function. Their research is incredibly exciting and we discuss all about it in today’s episode.

#08 Nir Barziali | Metformin, TAME Trial & Centenarian Longevity Research

Dr. Nir Barzilai is well-known in the field of longevity and beyond and is the director and chair of several institutes focused on Aging research. Just to name a few, he is the director of the Institute for Aging Research at the Albert Einstein College of Medicine and of the National Institutes of Health’s (NIH) Nathan Shock Centers of Excellence in the Basic Biology of Aging.

Dr. Barzilai’s research interests are in the biology and genetics of aging, so he has done extensive work studying centenarians and supercentenarians some of which he discusses in today’s episode and also in his book Age Later.

He is currently leading an international effort to approve drugs that can target aging. Targeting Aging with METformin (TAME) is a specific study designed to prove the concept that multi-morbidities of aging can be delayed by metformin, working with the FDA to approve this approach which will serve as a template for future efforts to delay aging and its diseases in humans.

In today's episode, we discuss centenarian longevity genes, what makes centenarians live so long and the role of growth hormones in longevity. We also dive into metformin's healthspan benefits for longevity, metformin & COVID, if metformin blunts muscle mass and causes hypoglycemia, and metformin dosage.

#06 Morgan Levine | Epigenetic Clocks, SystemsAge Clock and Epigenetic Reprogramming

Dr. Morgan Levine is an Assistant Professor of Pathology and Epidemiology at the Yale School of Medicine. Morgan runs the lab for Aging in Living Systems at Yale. Her labs works on Epigenetic Clocks, Epigenetic reprogramming, cellular aging trajectories, and in trying to understand the causes of Alzheimer’s disease.

I can’t even begin to tell you how much I learned in today’s conversation!

We extensively discussed Morgan’s work on developing a new Epigenetic clock called the SystemsAge clock, which is very cool and can test the biological age of different organs & systems in your body.

We also spoke of different features of these clocks, interventions tested, what we know so far on reversing your biological age, some of the limitations of the clocks, why she is so excited about epigenetic reprogramming, and some of her work around Alzheimer’s disease.

Most people are interested in knowing their biological age and epigenetic clocks are a way to test it. So I’m sure you will find this conversation on epigenetic clocks fascinating and walk away understanding them much better!

I hope you enjoy my conversation with Dr. Morgan Levine.

#05 Matt Kaeberlein | Aging Interventions - Rapamycin, mTOR, CR, Sirtuins, NAD+ & Dog Aging Project

Dr. Matt Kaeberlein is a Professor of Pathology, Adjunct Professor of Genome Sciences, and Adjunct Professor of Oral Health Sciences at the University of Washington. His research interests are focused on basic mechanisms of aging in order to facilitate translational interventions that promote healthspan and improve quality of life.

Matt has been around the field of aging for a long time and knows more about the work in the aging field than perhaps anyone else. This was a comprehensive conversation where we spoke about how Matt thinks of aging from a functional perspective, caloric restriction and aging, whether NAD+ precursors are effective in slowing aging, the role of sirtuins.

We discuss quite a bit on rapamycin and how it’s shown to be effective in improving the health of mice. Matt is very excited about the potential benefits of Rapamycin in reversing aging and he got me excited too. Matt is also working on the Dog Aging project and in trying to understand aging in dogs.

Finally, we end the conversation by talking a bit on what good scientific practices should look like. Matt approaches his work with an unbiased lens and that’s very respectable. Matt has a brilliant mind! I had such a blast talking to Matt and I hope you enjoy the conversation as much as I did!

#02 Dr. Jean Hebert | Brain Aging, Tissue Replacement, and Radical Strategy to Fight Aging (Potentially Forever!)

#02 Dr. Jean Hebert | Brain Aging, Tissue Replacement, and Radical Strategy to Fight Aging (Potentially Forever!)

Jean M. Hébert, PhD, is a Professor of Neuroscience and Genetics at the Albert Einstein College of Medicine in New York.

Dr. Hebert was fascinated from a young age by how a single cell forms the whole human body following an innate genetic program and he got excited by how knowledge of this process might be used to improve our biological fates and defeat aging.

He is a molecular geneticist by training and obtained his Ph.D. from the University of California, San Francisco, before specializing in the study of how neural stem cells form the brain at Stanford University.

His lab is now developing ways to use cell and tissue replacements to repair and rejuvenate the brain. More specifically, the Hébert Lab is engineering tissue that increasingly resembles normal neocortex, the part of our brain that performs our highest cognitive functions, and testing the ability of this tissue to integrate with the host brain.

Jean is the author of Replacing Aging.

We had an extremely interesting conversation ranging from why Dr. Hebert thinks the other methods to fight aging may not work if the goal is to beat aging, we discuss 3D organs and tissue cell replacement and we also discuss the neocortex and what it takes to replace one and lastly we talk about how plasticity in the brain works.

Even if you are not associated with the field of longevity, you’ll find this conversation interesting and refreshing.

#01 Reason, the Founder & CEO of Repair Biotechnologies | How to Fight Aging, NAD+ Boosters, Senolytics & Curing Atherosclerosis

Reason is the founder and CEO of Repair Biotechnologies. Repair Biotechnologies is a biotech company focused on developing therapies that treat medical conditions arising due to accumulation of excessive cholesterol. They have developed a unique Cholesterol Degrading Platform to reverse the damage that leads to heart disease and atherosclerosis. More than 5 million people die every year due to atherosclerosis and Repair Biotechnologies is trying to solve this problem.

Reason is perhaps most well known for his popular blog Fight Aging! where he distills the latest research in aging science. For anyone looking to stay informed on aging & longevity research, I highly recommend you checking it out - it’s one of the best resources out there.

In today’s conversation, we discuss how to think of aging simply, the differences between slowing and reversing aging, NAD+ boosters and their effectiveness, reversing aging using senolytics, how his company is working on a gene therapy to reverse artherosclerosis and cardiovascular disease, problems with Alzheimer's research, working towards aging rejuvenation therapies, how one can get involved in the aging space, and much more.

Note: We discuss a fair bit on cellular senescence and senolytic therapies. For those unfamiliar with the concept, cellular senescence is a process in which cells lose their ability to divide but refuse to die. When these senescent cells don’t get cleared away, they release harmful substances and damage the cells around them too.

It is well known that senescent cells accumulate with age, contributing to inflammation and aging itself. Senolytics is a class of drugs that can selectively clear away these senescent cells and they have shown promise in delaying and preventing age-related diseases.
